Is it best to check automatic transmission fluid while in park or neutral and does matter if vehicle is running not?

You should check your automatic transmission fluid with the engine running and the car in Park.

Checking it while the engine is off will give you an inaccurate reading because the fluid won't be at the correct operating temperature and pressure. The fluid level needs to be at its correct level while the transmission is at operating temperature.
