On a 1995 Ford E350 with a 5.8L engine (a 351 Windsor), the number one cylinder is located on the passenger side, at the front of the engine.
It's important to note that this is viewed from the front of the vehicle. You'll see the firing order stamped on the engine somewhere (usually on a valve cover or intake manifold) to be absolutely certain, but this is the general location.
